چکیده

The development and application of neuromelanin sensitive MR imaging has allowed the detection of significant changes in the substantia nigra (SN) of Parkinson’s disease (PD) patients, with high sensitivity and specificity in differentiating PD patients from non-PD aged controls, even in early disease stages, namely at the time of clinical diagnosis. These MR neuromelanin changes in the SN of PD patients reproduced in vivo long known characteristic pathological changes of PD. Several image evaluation methods have been used, corroborating the reproducibility of the data and enabling wider applications of this imaging technique in the clinical practice. In this review we analyze the background and the technical aspects of neuromelanin sensitive MR imaging, focusing on the applications of these specific sequences for the study of PD, emerging as a possible disease imaging biomarker and a promising tool for individual patient evaluation.

Residual substantia nigra neuromelanin in Parkinson’s disease is cross-linked to -synuclein

The pigmentation of substantia nigra pars compacta dopaminergic neurons is due to the presence of neuromelanin, an irregular macromolecular pigment belonging to the family of melanins. Depletion of neuromelanin in Parkinson’s disease is typically indicated by loss of brown color in this area. Unlike that from controls, the pigment extracted from substantia nigra of parkinsonian patients seems t...

Reduction of neuromelanin-positive nigral volume in patients with MSA, PSP and CBD.

OBJECTIVE Diseases presenting extrapyramidal symptoms are accompanied by nigral cell loss. In the previous study, we demonstrated the reduction of the neuromelanin-positive volume of substantia nigra (SN) pars compacta (SNc) in patients with Parkinson's disease (PD) using 3-Tesla MRI. In the present study we investigated the neuromelanin-positive SNc volume in patients with the other parkinsoni...

Brain Magnetic Resonance Imaging (MRI) as a Potential Biomarker for Parkinson’s Disease (PD)

Magnetic resonance imaging (MRI) has the potential to serve as a biomarker for Parkinson's disease (PD). However, the type or types of biomarker it could provide remain to be determined. At this time there is not sufficient sensitivity or specificity for MRI to serve as an early diagnostic biomarker, i.e., it is unproven in its ability to determine if a single individual is normal, has mild PD,...

Distribution of Iron in a Single Neuron of Patients with Parkinson’s Disease

Synchrotron radiation x-ray fluorescence (SRXRF) spectroscopy was applied to non-destructive elemental mapping in the melanized nigral neurons obtained from patients with Parkinson’s disease (PD) and a control subject. The cause of PD is unknown but many researchers consider that excessive accumulation of metallic elements (mainly iron) has a role in the generative process of PD. Microbeam imag...

A Comparison of Substantia Nigra T1 Hyperintensity in Parkinson's Disease Dementia, Alzheimer's Disease and Age-Matched Controls: Volumetric Analysis of Neuromelanin Imaging

OBJECTIVE Neuromelanin loss of substantia nigra (SN) can be visualized as a T1 signal reduction on T1-weighted high-resolution imaging. We investigated whether volumetric analysis of T1 hyperintensity for SN could be used to differentiate between Parkinson's disease dementia (PDD), Alzheimer's disease (AD) and age-matched controls. MATERIALS AND METHODS This retrospective study enrolled 10 pa...
